The historical Watling Street (established by the Romans), now termed the A5, passed through the nearby settlements of Hockliffe and Dunstable roughly two thousand years ago, and the Romans would have at the very least been aware of the village as they passed it.

The Romans were in turn followed by the Anglo-Saxons, and it was the Anglo-Saxons who set up a market in about the 10th Century, and antiquities discovered denote that they were settling the neighbourhood undoubtedly from the 6th Century. The commune as such was not referred to till 906 after the Danes signed a peace treaty at Tiddenfoot, the Domesday Book (1086) recognised Leighton Manor.

The parish of Leighton Buzzard, over the years, incorporated a considerably greater area compared to the town alone, and covered the hamlets of Heath & Reach, Billington, Stanbridge and Eggington. The "Leighton" in the name of the town appears to come from the old English for "a clearing in the woods", the "Buzzard" piece wasn't inserted before the twelfth century at which time the Dean of Lincoln had to distinguish it from the other "Leighton" that was inside his diocese. The Manors of Leighton were tenanted to an assortment of tenants through the generations, including the Leighs of Stoneleigh, who backed Charles I in the English Civil War (1642-1645).

Leighton Buzzard trainThe settlement carried on growing through the following years and the appearance of a railway line and a canal during the 19th Century saw the town evolve and develop.
